I have a 2000 F350 i purchased that already has a tune on it but no one knows what the tune is or where the tuner is. I have a friend who can flash it back to stock. But his computer or my tire size programmer can see the key to on position and do not start. is there a fuse or relay that can cause this? The truck runs and starts fine. Any help would be greatly appreciated.

What exactly are you saying/asking here?
If it was flashed with a plug-in-play flash tuner, you probably will not be able to read the tune. I would say your best bet would be to flash back to stock so you know where you stand. If you are planning to install a chip, it will override any tune flashed on the PCM. If your friend is willing to flash to stock, I would do that regardless, that way you know where you are starting from.

Are you saying that you get no power out of your OBDII connection? I've been where you're at with your tune, I'm 3rd owner and PCM is live tuned. I have kept it and am OK with it so far. If you connect a scanner and take for a run and run it through the paces some of the guys here can tell you a little about how it's tuned.

Very confusing post.
If the "programmer" won't cycle during the event its most probably not comparable with the current Tune, coorupt data stream, or similar. Or, require uninterrupted Power during the process.
If he's attempting to re-flash the PCM to "OEM Programming" the file may be corrupt.
Not addressing your question, but here's something to think about if you flash the PCM back to stock. If your truck has injectors that need a custom tune in order to run, you'll be cutting your left arm off if you flash the PCM back to stock.
