I just bought a 2000 Excursion V10 with 220,000 miles LOVE IT. It has been taken care of and everything but (of course there is a but...) the transmission does not like being in over drive. It seems to 'buck' not slip when under any load in over drive. I can accelarate and have it down shift with no porblems then it is fine.
Also the over drive light on the shifter will not come on. I pulled the button and put a tester to shifter but no power was going to it. (I had the motor on in park with the passenger door open.)
Could these be related?
A good place to start would to check the wiring going to the shifter. A very common problem is nicked/frayed wiring, especially where the wire comes in contact with the metal of the shifter. Remove your steering column covers and check it there. Look it up on here and there are numerous threads with pics. As for the bucking when in overdrive, mine was doing it when I had a bad COP. Awsome...I will be looking at those tonight. I was getting a mis-fire in #2 so I will start with that one. i found a great write up on how to check the COP. Also I will be pulling the cover off the column and looking for frayed wires.

Do you have a link to the COP testing write up? This is on my to-do list as well.
What you have is a spark plug mis-fire. If you replace the plugs and the plug boots you will solve the problem more than likely. Do this first! You are looking at about 90.00 in parts. Now if thats not the problem it leaves the COP. But, more than likely its not a COP.
