Depending on why you want to lock up the TC it's really not a good idea to do so unless your gonna beef up your tranny. Billet input shaft, heavier duty clutches etc etc. You can get a tune that'll lock it up through all the gears but beware, you might get more than what you've bargined for. JMHO.

I thought locking your TC reduces the amount of heat generated. Since heat is the number one cause of transmission failure I would think it would be a good thing. What are the negative impacts on locking the TC?

I'm no expert on trannies and maybe someone will chime in with better info but from what I've gathered by reading and hearsay if you think about it locking your TC up through all the gears puts an enormous amount of strain on a stock tranny. In my head and correct me if I'm wrong but your locking the shafts together now that said your trying to get your 8000# truck and whatever else you got behind it up to your cruising speed with no slipping of the TC. That's alot of strain on that shaft. This is just my outlook on it so don't qoute me on this. I've just heard of guys running a race tune that locks the TC through all the gears and using it as a daily driver and they end up burning up their tranny. Just my 2cents.
