I wanted to see if we could figure this out but now the truck in at the shop today!!
At first I might think it is something simple like a U-joint(most likely the one closest to the rear axle), since 4wd makes it quieter-due to the fact that now the front axle is taking up the slack that is causing the "noise" from the back.
More info would be nice, and help. Like, year of truck, F150-F250 etc. Mileage, transmission type(auto/standard)
Driveline noise can be tricky, however if you tell us what noises you hear on acceleration(light and hard) and de-acceleration(engine load) may help.
Since your truck is at the shop now, perhaps let us know what they found. I hope you don't get ripped off, or mis-diagnosed. I have heard of people having their rear axle completely rebuilt(around $1300) then find out it was a transmission mount, that cost $150.
Some areas of this forum get alot of attention, and quick responses, some don't. At least you posted yours in the right area. Altho-this section doesn't get quick responses normally, it seems.

My first post was on Nov 3rd..I think I posted in the 2004-2008F150 thread..I only recieved one response and it was from Tylus who told me to "search the site for the term IWE" which I did..and it sounds like this is our problem...but I had some more questions that I posted under my first post and nobody helped me, so I tried to move my thread to this thread...that is why there is so much "junk" in my first post..(which is what "Stuart" just posted about here) anyways...after posting here..I still heard nothing..
so..this is the problem...we have a used 2004 F150 4x4 that has 30,000 miles on it..we have had it almost 4 months. while I was driving the truck in 2wd, it made an aweful sound, like a banging/thumping/rattling...I stopped the truck and looked under it b/c I thought I ran over something or something was dragging...nothing was there...started down the road again and it sounded like it was grinding....when I put it in 4Hi the noise is not there...the noise is only while it's in 2wd going forward or reverse, the noise is there.
we thought we would start with the IWE..so I went to our ford dealership and bought the "new updated" part that has the shield on it (to keep the water out) and the noise is still there.
so now the truck is at another ford dealership (being ours here in town just closed it's doors today after 93 years) I will hear today what they find...hopefully its not to bad...they said it could be the hubs or something.
