My f100s lighter wasn’t hooked up, as a result I have a couple questions
1. What color/number of wires are needed to make it work? The wiring behind radio and etc is butchered and replaced, but maybe the correct wires are still there.
2. How do I read the voltage? This depends upon how many wires are involved of course, if it’s only a positive I assume I just touch the negative lead of voltmeter to the dash or similar? If it has a ground wire, where is it supposed to attach on both ends?
If you can't find the wire described above, you can add your own 12v wire from the battery, ignition, or the yellow 3 socket ignition source over near the headlight switch
There should be a green/yellow stripe wire going to a yellow 3 (male to female) Y shaped pin receiver behind the glove box, look up under and to the left....it also powers the glove box light if you have that. Should have one wire plugged in there and then it attaches to the back of the cig lighter stud.
Gets ground when installed in dash. The yellow Y block looks just like the 12V keyed connector behinds the cluster.
The yellow Y you are looking for is NOT 12 V keyed, but 12V HOT all the time.

3-pin female Y-connector to the left of the cluster is 12-V Keyed source?
3-pin female Y-connector to the left of the glove box is 12-V IGNITION source/ Hot all the time?
As far as I understand, yes the one behind the cluster is keyed 12V ='s (tun on the ignition key in the 1st position to the right and you get 12V power). And I understand the one behind the glove box is 12V HOT all the time. For glove box light, cig lighter.
Power from battery to the headlight switch (to turn on dome light), dome light door switches, bed light switch (B pillar mounted), under dash courtesy lights.
