Yesterday I went to start my 2016 6.2 with 4500 miles on it and it had a very loud knocking. This was not the startup tick that some 6.2's have, I have heard that before so I know it wasn't that. The knocking lasted long enough for me to get out of the truck and listen to both banks of the motor and get back into the truck. When I brought the rpm's up slightly the knock finally went away. This was a very loud internal type of noise. I started it today and it was fine, nice and quiet. It has never had any kind of noise like this. I got the oil changed by the dealer about 500 miles ago and my first thought was the oil filter anti drain back valve might have not worked properly. Should I be worried about damage? could this just be a isolated issue? Not sure what to do!!!!
The start up tick is normal, but not a knock. This is the first post I've seen about any 6.2 knocking. Don't know if any dealer can/will do anything about it unless they can hear it happen.
If the it went away it wasn't the bottom end. More than likely a lifter bled down and it took a little bit to pump back up. Was the truck being driven daily or had it been setting for a while?

This is were the oil is lost and pressure has to be built back up.
^^^^ correct, it has been covered several times here.
My Ford has done this on occasion and my previous truck with a similar type of engine. You are most likely hearing chain and valve noise until the oil pressure builds.
The truck had only sat for 48 hours, it has ticked on startup a couple of times, but this was a much deeper more pronounced sound. I have thrown rods before and it reminded me of that noise. Plus I didn't say this before, but it was running really rough until the noise went away.

If it happens again get out and look at the belt, you can see it through the wheel well. Mine would make a knocking noise when it was cold and the noise was in time with the belt jumping up and down, when away after warm up and stopped all together after about 10 or 15K miles.
OK let's go a little deeper. How long have you had the truck? How old is the fuel? What was the temperature when this happened?
The reason I ask these questions is with only 4500 miles the engine is still tight and if it was cold and the fuel was a little old it could have gummed up a valve or two causing them to stick and make the noise and the engine to run rough.
Take it in and have it checked. Mine has never made a knocking noise. A slight occasional light ticking noise that disappears in seconds but not a knocking sound.
I just changed my oil to Castrol High mileage 5W-20 even though I have only 20,000 miles on the truck and the occasional ticking noise went away. The oil pressures appears to be a bit higher than after previous oil changes and the engine is quieter than it has been since it was new. It might be worth a try for anyone else is experiencing the occasional ticking noise.
