Yes. That is the main connection that controls the transmission. Without it, it will default to 4th gear.
If you put a new E4OD, that is your problem. They changed the connector in '95. Your choices at this point are:
1. Swap in an older transmission
2. Change out the solenoid pack to one from an older transmission
3. Cut the connector off your truck and splice on the new style connector.

Well it sounds like there are only two different ones, 94 and down and 95 and up. Sounds to me like your truck has a '94 plug and the trans has a 95 plug.
You could always snap a pic of the plugs on your truck/trans to compare when you hit the parts store.

The E4OD is actually a C6 with overdrive and electronically controlled. If I were replacing an E4OD I'd just take the hit and find a reputable tranny shop to upgrade it and make it more durable. Actually I did do that exact thing with the 700r4 in my '84 K5 blazer. A built E4OD will cost more but will last as long as anything.

Not sure why everybody is jumping on the 94-95 plug change for the diodes.
If you have a 1994 and you got a transmission for/from a 1994 then you will have the right plug on your harness and transmission.
The problem with that plug is the heat from the exhaust causes problems right where the wires go into the back of the plug and the wires brake off. You should have a heat shield bolted to the transmission at that point.
