checking Transmission fluid level 6R140

Probably a simple question for someone who knows transmissions. I looked at the manual and it's not entirely clear. Should the transmission fluid be checked with the truck running? I know from the manual that it says check fluid on a level surface and when transmission is warm, it's just a bit unclear about running vs not running.

X2 running in park hot on level ground.
Filter and fluid should be changed every 30K less if you tow heavy.
