Now that's a loaded question for our trucks!
In my case, I pulled the engine and gave it a full rebuild just because I had to remove a leaking heater core and figured I'd do them both.
So I'm not much help if you don't want to remove the engine to make it easier. Luckily, it can be done without it.
I think there are some threads here with pictures and full details on the little tips n tricks for making it easier. I'm not sure where the are so can't give a link, but I'm sure someone will.

With or without a/c? GIANT black box in engine compartment on the passenger side ='s a/c? If you do not have a/c...look up under the dash behind the glove box. See that black box (L/H diagram), that has to come out and be opened up to access the heater core. With a/c same thing just a giant PITA IMO. Once you tell us with or without a/c the answers will roll in.
