I have done it a few times...I believe .you need to drain the Rad first.......then disconnect each line .....take out the Battery.....( I think ) . remembering where everything goes.....maybe take pictures ......save the coolant.....Then you need to take the Box off from the Engine bay ......IIIRC Blower Fan First...then box as a unit ...Heater Core is what is left ....Most nuts are Inside the Cab ...they are nuts with a Flange..... RE and RE the Core and Re assemble.... make sure that core is attached solid ......and it is wise to replace the thermostat as well ......It is not a big deal ....Do not Overtighten the clamps....

Blageurt, it is much simpler on the 88 e150s.
1- Might want to drain some coolant so it will be below the heater core level.
2- Remove air filter box.
3- Remove the hoses from the heater core under the hood.
4- Remove the 2 screws holding the modesty panel on under the dash on the passenger side
5- Remove the 4 screws holding the heater core cover on the left side of the heater core case under the dash.
6- Remove the screws and retaining bracket at the bottom of the heater core.
7- Remove the heater core and seal from the case.
8- reinstall in reverse order.

That's ok! My mom's 78 E100 has to be done the way you described. I just happened to have already checked on my heater core. It is not leaking yet (I don't think) but I am going to replace it and the hoses in the next few months just as preventive maintenance since they are 30 years old. That also gives me time to search around and find a OEM Ford heater core at a cheep price. Procrastinating can be an advantage sometimes!
