Intercooler or transmission cooler?

I haven't done any heavy hauling with my f250 since I've owned it. Well today I was helping my inlaws move pulling a 16 foot trailer loaded down. Anyways had my overdrive off and running 55 mph at 2700 rpm I hear what I'm guessing is the intercooler or transmission cooler? My next question is it always this loud?

Yep, normal. You didn't have to have the OD off if it pulls without shifting between gears.

Why?

Because that's how you drive when you tow.

Rule of thumb is, take OD off whenever the trans is hunting for the right gear, when headed up a grade, towing heavy.
