Rear wiper and defogger aren't working on my Expy. Have checked all fuses and relays in both dash and engine compartment, all are good, and have replaced the wiper motor. Is there a junction point for the wiring going to the tailgate that I can begin my search for loose connections?
The two circuits have only 2 things in common. They both pass through a 60-pin connector down in the driver's kick panel, and they both depend on the liftgate being bonded to the main body for a ground. I would first investigate a missing or broken ground to the lift gate. You can make a temporary jumper just to see if you can get it working. Keep in mind that the rear wiper is disabled if the rear hatch or glass are popped open.
I'll check those tomorrow. I hadn't thought about a ground issue, thinking that there would be sufficient contact through the hinges to accomplish that, but I'll try that, and check under the kick panel if a new ground doesn't solve things.
The 3rd brake light is working ok though. Does it ground through the lift gate as well, or through it's harness?
Added a ground wire between the liftgate and body, still not working. Also checked the connectors behind the driver's kickplate. Nothing looks amiss, but I don't have the means to check everything there.
I'm running out of things to check. I do know that the vehicle is correctly determining that the door and glass are closed; when closed and the rear wiper is turned on, the [wiper up] and [wiper down] relays in the engine compartment power box operate, when opened, they do not.

Below is a pin-out of the connector in the driver's kick plate. You'll probably have to reverse this in your head since you'll be looking at the back of the connector and not the face of it.
Pin 4 is white/orange and pin 5 is black/light blue, these run back to the rear wiper motor. When you turn the wiper on positive and negative are one way, but when you turn the wiper off both wires switch polarity to reverse the direction of the wiper motor.
For the rear window defroster pin 39 is brown/light blue and it goes positive when it's on then goes dead when you turn it off.
You never mentioned it, but does your defrost switch indicator light turn on when you press the switch on?

The motor has 4 wires going to it, wasn't sure what each one was, until I found a wiring schematic:
white/orange - Wiper Down
black/lt blue - Wiper Up
purple/yellow - Hi/Lo Park Sense
black - ground
So it looks like it's not dependent on the liftgate being grounded.
I have no power at the kick panel connector on #4, #5, or #39. I got power on #37 and #38(whatever they are) to verify that my test light setup was sound.
In that case, since there's no power on any of those pins, the wiring from that 60-pin connector on back is good. Perhaps you have the infamous windshield leak into the GEM and/or fuse box.
Is there any chance these are two separate problems that happened at two separate times, and you only noticed them at the same time?
