Get access to a complete donor truck if you want to go the OEM route. That way you have all the wiring and sensors you will need. Actually thats the complicated part of the whole thing.
For factory original electronics you will need signals from:
FILP sensor or Throttle position sensor located on the driver's side of the injector pump.
MLPS or manual lever position sensor. This tells the transmission where the shifter lever is and also has the neutral safety switch and back up light switch built in.
VSS of vehicle speed sensor. If your truck has factory speed control, it will have this sensor at the transmission end of the speedometer cable. Same for the E4OD.
You need to have a tachometer sensor on the injector pump drive gear houseing (big 1" nut with 2 black wires comming out of it).
The brake lights will also have to be wired into the computer.
There is also a barrometric sensor in there some where to adjust for different altitudes.
What I did instead was use an aftermarket computer. Its programmable and has the ability to switch shift tables on the fly. Programming software is free, and you can use a laptop or desktop PC for tuning/diagnostics. Signals are required from only:
VSS, MLPS, and TPS.....and thats it. You can order the wiring harness with the computer and its 80% plut & play. If you can afford the $650 cost of the baumann system, I highly recommend it. I've been using mine for a few years now and I never had a single problem with it. Its one of those products that simply performs as claimed.
The bolt pattern is the same, but the flex plate offset from the mating face is different between the E4OD and the C6. I made a 1/8" aluminum plate to take up the difference, wasn't hard. The other option is to swap over an E4OD type flywheel.
The driveshaft will need to be shortened or replaced depending on what is cheaper for you.
The C6 cross member could probably be moved without having to be replaced.
The shift rod that connects the truck to the transmission will need to be replaced or lengthened. The rest of the linkage works no problem and the shift quadrant lines up nicely.
Do yourself a favor and buy a NEW aftermarket cooler as big as you can find and install a trans temp gauge with the sensor on the outlet line of the transmission (front line).
If you rack up a lot of miles on this truck this is a good upgrade that will pay off in a few years, but don't cut corners and consider rebuilding and upgrading the transmission before putting it in. Build it as high as possible if you plan to turbocharge and tow a lot.
Sterling axles like my 1986 have the casting for the sensor in the housing, but no actual sensor is there.

If you are asking about swapping within E4OD transmissions to put the cable drive for the speedometer, the answer is yes but....
You will probably also need to swap the tail shaft out of the old tranny to the new one. Some post 1992 E4ODs had the worm drive still on the tailshaft even though the cable drive was eliminated because of left over parts. Swapping the tailshaft involves a complete teardown of the transmission since the tailshaft is nearly the very last thing to come out (2nd last if I remember right).
If your truck is 4wd or ever want it to be, then just look for a 4wd transmission and pop an older transfer case on it that has the cable drive.
