Fuel drain back
I have an 88' 300 six efi that you have to let the fuel pump run for about 10 -15 sec. before you get a reliable start. New fuel pressure reg. I understand the fuel pump has a check valve in it. Most likely culprit?

Yes. That is your most likely culprit. If the truck runs fine otherwise, I would consider installing an in-line check valve on either side of the pump. May be better to install one on the suction side vice discharge. There is a similar fix to a similar problem on the Jeep 4.0 Cherokees, etc. that you can probably do a search for and find.
