My 96 F250 has a 460 and E4OD so yes they will bolt together just fine.
C6 is a 3 speed and the E4OD is a 4 speed, the OD means overdrive. The E means electronic, so it is shifted by a computer.

Thanks for the info, When i asked for the info about the OD and the C6 I was asking if the E4OD could handle the extra power from a 460, but if yours has it then i guess so! Thanks alot for your help.
It depends on which E4OD you have--there are several versions. The 300, 302 and 351 all have one bellhousing bolt pattern, the 460 has a different one (I don't recall if it matches the diesels or not). So there's at least two, maybe 3 (I'm not real familiar with the diesels and the 460, so what I know, I read here).
The C6 is of course, legendary in reliability. The E4OD is less so, but it was also put behind much more powerful engines--the PowerStroke and a newer 460. Later versions of the E4OD are considered to be much better than the initial release.
Gotcha, Thanks for the info. I guess if anyone ever wanted to put a 460 in a 90's bronco, there best bet would be to go find a 78 bronco and use the trans transfer case and axles.
