A set of good gauges would be an added benifit, for you to monitor the EGT, Boost pressure, and Trans temp while towing. I towed my toyhauler 11K# without any gauges in my 03 and early on with my 05, but frankly don't know how I got along without them. I feel much better being able to monitor what is going on while towing that heavy of a load.
I'm surprised that no one mentioned axle ratios yet. I tow a 10,000lb trailer with my 07 powerstroked E350. I ordered it with a 410 rear end as that's what Ford recommended for a 10,000lb load. I get 20mpg not towing and about 14 towing but rarely drive over 70mph with the low rear end. On the other hand, the 355 ratio might get you some better mileage not towing. However, it's not recommended to use a 355 when towing over 8,000 lbs.
FOSE88, we have, just not in this thread. A 4.10 should be standard in the 250 and 4.30 standard in the 350. 3.73 is the only current option you have from Ford, unless you go aftermarket.

One other thing to remember about the "E" vans with a 6.0 and the "F" trucks with a 6.0. The "E" vans get a detuned motor that does not put out nearly as much Hp & Torque as the 6.0 in the "F" series trucks. This is largely do to the inadequate cooling available on an "E" series van. Therefore the "E" series vans need a lower gear ratio to pull the same weight as the "F" series trucks.
"E" series 6.0 = 230Hp & 440lb/ft
"F" series 6.0 = 325Hp & 570lb/ft
