I have a 2000 expy with a 5.4 and a 4r100. I have noticed searching through many posts that it is a good idea to replace the fluid every 30k. What fluid do you guys have the best luck with?

Your call on the AMSoil. It is a dual rated fluid and Ford states clearly in the manual NOT to use dual rated fluid.
Well, I looked high and low for a straight Mercon trans fluid the other week. (o.k., about 4 places). Then I went to Ford. Guess what, they don't have it either, they only have a Mercon dual rated fluid (not Mercon V).
On another side note, about three parts guys said to use Mercon V. Know who else said Mercon V??? Good old toilet paper Haynes Manual at Pep Boys did, right there in their table. Laughed. Just one other reason I buy Factory Service Manuals.

For Mercon applications, use Mercon III / Dexron
For Mercon V applications, use ONLY Mercon V
DO NOT use any ATF (like Amsoil) that says Mercon / Dexron / Mercon V

Seems to me that A Mercon/Dexron III/Mercon V would be a triple application. As I recall, one of the differences between Mercon and Mercon V is that Mercon V is synthetic. Seems to me that you should stay away from an Amsoil or other synthetic aftermarket performance blends then...
Tell you what, this whole trans fluid is incredible how much a pain it can be, but sooooo critical to the performance of your trans. Not like the engine oil - running an oil change with a slightly different viscosity because you happened to not have 5W20, and mix a little 10W20 of even a different brand in the crankcase. Went through a ton of pain on a chrysler transmission when even though the documentation said Dexron III was o.k. to use instead of the Chrysler fluid...
Many threads AND Ford TSBs say DO NOT MIX Mercon III and V. They are not compatable. Basic rule is, what the dip stick says, use that. The internal specs are designed for the recommended fluid and III and V are different. Even Ford dealers mess it up. I had to show a local service manager the TSB as he wanted to put V in all trannys. He screwed up one of mine and changed the fluid again that day for free. Paid for III, got V. A cost savings but wish it hadn't happened. I have a 97 Expy (Mercon III) and a 98 Mountaineer 5 speed (Mercon V) which is the one they screwes up.

Mercon V is a synthetic blend, NOT full synthetic!!
Two years ago I bought a used 2002 F-350 SRW 7.3 diesel with 36K miles on it. Shortly there after I took it on vacation with the standard lube that was in it...(tranny & engine). I averaged 17.8 MPG on the trip. When I returned, I changed all of the fluids with Amsoil products....Amsoil ATF, Series 3000 Diesel Oil, and Sythetic diff/gear oil. 25K miles later on the next trip I took, I averaged 19.6 MPG.....same trip, same route, same load. I was pleasantly surprised to say the least! Hope this helps.
