I took the drive shaft off recently and had it balanced and checked out. Everything is perfect on it. While I had it off, I tried to move the flange coming from the rear end.
With the tires on the ground and not moving, I was able to move it back and forth about 1/16 of a turn. I thought this was too much play. Should it have this much play and is there a way to adjust it.
The reason I ask is because the truck vibrates at around 65 and will clunk when you let of and then step back on the gas. It thought this was coming from the drive shaft but it turned out to be fine.

I had a similar problem with the clunking at take off and coming to a complete stop. I have the 8.8 traction lock rear and what I did was replaced the gear oil and lube up the slip joint on the rear driveshaft. As for the vibration check and see if any weights were knocked off on the driveshaft or you may just need your tires balanced.
The tires and the drives shaft are balanced. I will try replacing the diferential fluid though.
Does anyone know if I have too much play in the differential or how to tighten it up?

There should be a little play in it. You can get the specs out of a Haynes manual for how much backlash or play should be in the diff. You'll need a dial indicator to check the gear backlash. The only way to check is with the diff cover off and you check it on the ring gear. The only way to adjust is with shims. There is also a special tool from Ford called a pinion depth gauge that you'll need to set the depth on the pinion gear.
It's rather difficult to set up a rear end properly.
