Most areas have a measured mile on the high way or a side road that the towns use to calibrate Taxi meters, ask your local weights and measures where it is and check your odometer against the measured mile.
Basically what you are wanting to do is compare mpg with a ratio of 1:1 to the overdrive ratio. This is easy. After the trans swap is complete take a trip, only going into 4th gear, calculate mpg then take a trip with overdrive, calculate mpg, and that is your comparison. This eliminates any other variable, providing the most accurate comparison since it will be the exact same drivetrain.

First tank full with the overdrive is looking like 10.5 MPG. My reciepts from the 4 speed blew out of the window so I'll have to make a run without using the OD.
