300/302/351W all use the same bellhousing pattern. Flywheel/flexplate is another story.
300 is internally balanced, and the rest are externally balanced, so the flywheel for a V8 will have a counterweight, the I6 flywheel won't. so yes a AOD or the later 4R70W electronic version of the aod with better gear ratios will fit
Check here for part numbers for the flexplate and some "info"

Try over on the ford six forum Im a member there as well but I dont know as much about the big six 240/300 but i can get you in touch with the guys there who know if you have any technical questions i know who to ask.
if you were talking the ford small i6 (144,170,200,250 or the australian ford 221 or crossflow 250) then I can be of assistance as i have run all of those at some point.
got a 200 right now in a 65 mustang with a rollerized c4 transmission good to 800hp with a mild stall. the head is a custom aluminum intake with plenum and rare two barrel vintage holley made for the austrlian 4.1l I6 with a gt35r tubro and im looking to get 835hp from it.

That highway 4 speed (The SROD I'm assuming) is a piece of junk, but I'd put an NP435 in its place. It'd be a direct swap and less of a hassle.
The NP435 is an absolute tank.
Secondly, the 300 + C6 is a very robust combo, but the C6 is a power suck, and the 300 would be very sluggish behind it.
