Are you talking about the little plugs, or the big ones on the end of the rails? If talking about the big end plugs, then yes, I'd pull them while the engine is out. Replace the o-rings, or go ahead and replace the o-ring and plug if you'd rather.
Very difficult to get a ratchet and leverage on those while it's in the truck.
It should have the updated ones in it unlike the early PSD 94.5-97 but if you have the engine out it is definitely easier then later. I have not replaced one on the newer 7.3's in the 15 years of wrenching I have done. If they are really tight sometimes a little heat on cylinder head at the plug will loosen the locktite on the threads.
