First, and most important! You DO NOT want to disassemble the gearbox to replace the seal! If you do, you will regret it very quickly. These boxes can be a royal pain to reassemble correctly.
That being said, yes, you can replace the seal. I don't know if it can be done with the box in place or not. I've always done it with the box out of the truck.
Here's how to replace the seal. The "seal" that you actually see isn't a seal per se. It's a dust boot. To remove this take a sharpened center punch or something similar and pierce the seal and pry it out. This will expose the "C" clip that retains the actual seal. Remove the "C" clip then remove the seal the same way you removed the dust boot. Sometimes a spacer is used, replace it in the same position.
When I install the new seals, I coat the inside lip of the seal with a light coating of grease and I coat the OD of the seal with #2 Permatex non-hardening sealer.
Use a deep socket to install the new seals. Don't forget to install the spacer (if used) and the "C" clip.
