What are we talking about? lifters(located at camshaft)pushrods or rocker arms(located under valve covers) one end of pushrod can be seen under valve cover the other end is in lifter which is in lifter valley under intake manifold. We are talking V8 and most likley hydraulic lifters?
Whoops swapped my parts up... Lifter will have excessive wear, or will not rotate in its bore. Rocker arms will simply be broken. (I wouldn't bet on rocker arms being broken.
If a lifter is gone, the cam more than likely is too.
if a lifter is malfunctioning the valve it controls may also be malfuntioning, you could take reading with a vacuum guage( they are cheap at the parts store and if you dont have one you should get one they are invaluable for tuning and diagnosis). you would have erratic readings indicating a valve problem otherwise it should read steady.
if this reads normal my next step would be checking the oil, not just the level but if its pretty dark might as well change it so run some sea-foam in it to degunk everything and change the oil, if its fairly good try adding some lucas oil to quiet that lifter down.
depending on your engine you may be able to see your lifters. on a 302 you can see them but on a FE i dont think you can so if you have a 302 you could pull the pushrods and check them and also see if one of the lifters has broken somehow or collapsed, this can be done pretty quickly. mine had a worn ball end on one pushrod it was more eggshaped/pointy than spherical.
also check your exhaust real good for any leaks i am hard of hearing in one ear and sometimes cant tell the difference at first since both issues producing ticking noises.

most of the time a visual inspection will tell alot, pull the valve covers and start, look for unusual valve train movement or lack of. if oil return passages are clear it should not be too messy,you can also use cardboard to contain any splashing.
Have a miss and a fast repeating and kind of quiet back fire already eliminated electrical and fuel for the miss

Like numbers said (lol) get a vacuum gauge. I'm actually willing to bet you have a vacuum leak somewhere and if the gauge reads low vacuum (closer to atmospheric pressure), this would be confirmed and it may actually be an easy fix.

Sounds more like a dead lifter or wiped cam lobe with the description of backfiring.
It is 460cid with a eldelbrock 750 carb on top of eldelbrock performer series intake manifold also eldelbrock cam and lifters with matching timing set . Approximately 110 000 mi since rebuild in 2004. Lifter always made s little noise since day one . My only form of transportation so didn't want to tear into it to far till I know for sure the problem
