I have only had the truck for a short while and its only been on the road for the last 2-3 days. I noticed tonight when I let off the throttle there is this clunking sound, and it also happens when I re-apply the throttle but not as bad. I was wondering if this may be a bad tranny mount or something like that? Any ideas?
BTW its a 1977 F250 4x4 with 35 inch tires.

Motor mount, tranny mount, universal joint, ring & pinion......
Check the u-joints first. They're easy enough to grab hold of by hand and check for slop. The rest (unless extremely obvious) might be easier with a passenger listening and looking to pin point the location while you operate the truck and reproduce the "clunk".
