I have a 1971 F100 that originally came with a 360 and automatic, it now has a 351W and C6. This may sound strange, but when I got the truck about 4 years ago, the speedo did not work, but started working a few months later and worked until I parked it do to gas prices about a year ago. Well I put it back on the road today and the speedo does not work again. Any suggestions what it could be, and if I need a new cable, what do I tell the twits at the parts store I need if its not a factory cable do to the new motor / trans.
From what you have written I don't think it is the cable that is the problem. I would think it is the speedometer head itself is the problem. But to be sure you can remove the speedo cable and inspect it, if the cable and the plastic gear that goes into the transmission are good then it is the speedo head. If it is for sure the speedo cable, just take it out messure it, the guys at the parts store should be able to get it from that. I don't think it matters that its a different transmision.

Broken speedometer? It could be that the speedo cable core has broken.
You can buy an internal speedometer core. It's a universal kit. You just remove the old core from the housing, cut the new core to size and re-install it in the speedo cable housing. Make sure to use white grease on the core. When the core runs dry, it binds in the housing and snaps.
However, before replacing the speedo cable core, look at the driven gear in the trans at the end of the cable. It might be stripped. The gears are still available from Ford.
