Short answer - yes you can. I'm not sure if the check engine light would be on or not, though. If you really want to do it right, all you need is a new computer from a junkyard or ebay that was meant to run your engine without controlling the tranny.
As a matter of fact I just rebuilt the 351W and then the transmission craps out on me! Will the C6 be a direct fit? Will I have to change my transfer case as well? and where will I get a vacume line for the Modulater valve?
A C6 from a 300, 302, or 351 will be a direct fit. Don't get one from a 460 or a diesel, it won't work.
Your transfer case will be fine. Vacuum lines can be bought anywhere. Since the C6 is shorter than the E4OD, you'll have to shorten/lengthen driveshafts and get a C6 crossmember for your truck as well.
Why not rebuild the E4OD? Make sure they use the updated parts and it should hold up to whatever you throw at it. Any GOOD transmission shop should be able to rebuild it for a lot less than swapping in a new trans.
All the local shops want $2000.00 to rebuild, And thats the cheaper ones, I can get a remanufactured one for $2200.00.

I believe there are several tools that you need to rebuild an E4OD. It's a job that I'm not sure I'd want to tackle - what if you got it back together and it didn't work? Or failed in 5000 miles? I'd be..... well, I'd be upset.
