Does anyone know if the auto/man locking hubs can still be used to engage 4 wheel drive if the ESOF vacuum pump (that normally eagages the hubs) fails? Is the manual setting a failsafe backup for the ESOF?

"Yes" to both of your questions. But, remember that the transfer case gears are engaged by an electric motor in the transfer case, so your dashboard selector switch must *also* be used to engage/disengage 4WD.
You probably already know that running your rig in 2WD with the hubs manually locked can cause excessive wear, so keep that in mind.

In theory, there's nothing to prevent you from modifying your vehicle to use vacuum-operated hubs with the electrically-operated transfer case.
As a matter of practicality, you should probably forget the notion. The sheer cost of replacing all those manual components with the ones needed to make the conversion will no doubt change your mind.
You maight also consider that many have chosen to replace their factory-installed shift-on-the-fly automatic system for the front hubs with the ~more reliable~ (fact) manual hubs.
It's your choice, though. One person's headache can be another person's passion.
