Do you hear the pump run when you turn on the key? It should run for a second or so and then shut off if you don't crank it. This is run by the PCM (Computer). This primes the fuel system for starting. Once running the pump is contolled by the oil pressure switch (I think- have to check the diagram- this is how GM does on some). Anyway, without the computer sensing that the engine has started, it won't run the pump. I believe this may be your problem. You may be missing this 'prime shot' so it won't pick up and let the pump run. When you manually prime it, it fires, the computer senses that it's running, and allows the pump to run.
My nephew had a similar problem on a Ranger, replaced fuel pump, had me test fuel relay-ok, and finally found computer was bad. This is what I'd look at.
