TKO 600 could work, depending on what type of truck you're trying to build. That's a street car transmission, so the gears are designed more for a car than a truck. If it's a 2wd truck that you want to be a freeway flyer (and not use it for truck stuff), then it could work.
If you want to use the truck for carrying or towing, you'd probably be better suited with a truck manual, like an NV4500, ZF5, or Tremec TR4050 (which is now offered with a 2wd tail housing), all of which are truck geared but have overdrive.
Easiest way to swap would be pick up a manual parts truck, so you can pillage things out of it like the pedal assembly, mounts, clutch linkage etc.
