Where to fill diff. from?

Guess its a dumb question, but i should fill from the top right? And not the bolt hole in the diff cover? (2000 E350)

From the top? You mean the vent tube?

These get filled through the oil level setting hole.

That's a fill plug, not a bolt hole.

Also check it for metal since it is magnetic when you pull out that plug and clean it. If you want to overfill park on a decline.
