I recently bought a 79 f350 4x4 regular cab with the 351. The guy who had it before removed both tanks and some of the lines. I have the side tank all cleaned out and ready to go back in .My question is if I can just run the line from the fuel pump to just the one tank? Does it need to have both tanks? Thank you in advance!

You don't need both gas tanks. My 77 F-150 had two tanks, the rear (aft) tank between the frame and the OEM in cab tank that was behind the seat. I recently removed the in cab tank, didn't need it, it created allot of noise anytime the fuel level was at half or lower with the float and could hear the gas swashing around (no baffles), plus I didn't like the faint smell fumes inside the cab and just didn't like the idea of a fuel tank so close to occupants. I drive our 77 F-150 between 900 and 1,200 miles annually, so I really didn't need both tanks. I had a gas diverter value underneath the driver's side of cab that I removed. It was electrically controlled and diverted gas from respective tank via the switch on the dash. So in short, you don't need both tanks.
