Are you cycling the key for 20 seconds about 6 times or more. When you bleed off the simple sep in the rail, air is not your friend. Try a repetitious cycle and look for leaks. Takes about 6 instances for me to get the air out when I pull my drain valve on the frame rail.
Did you use motorcraft filters???!!!

theres going to be a lot of air in the injector fuel lines, the fuel rail, the fuel cooler, and both fuel filters.
do the cycling thing before you try to start it.
also, depending on the year 6.4, there's a fuel cooler bleed air valve that looks like a tire air fitting. once you turn the key and hear the pump whine.....dont try to start it...leave the key in the on position (but dont turn it to start it) and go out and while the pump is running, bleed the air out of the fuel cooler using the fitting.
then do the 6 cycles of on and off for 20 seconds...before trying to start it.
