I'm using a recommendation from Jarrett, witches brew. 1 ounce of Kleen diesel per 4 gallons and 1 ounce of 2 stroke per gallon. Seems to have quieted her down a bit.

I had a 1978 E-350 back in the day and loved the duallies. 18 year old kid, knowing jack-**** about cars.. but I did learn you could do 1,400 miles on a flat, NP. Chicks loved it too.
I have never used any additives but 2 stroke oil to my diesel fuel on my tractor, just an oz per tank
I also use 2 stroke oil in anything carbureted for past ten years. E15 fuel once it evaporates, leaves alcohol with water in it. it corrodes aluminum fast if there is nothing to protect it.
I use two... The first is the Cetane Booster you can buy at your local Ford dealership. The other is Power Service Diesel Kleen. Both work well, and the truck runs a bit better when running them. I do it more-so for peace of mind, to ensure my injectors are happy.
I have run 2-stroke in the past. Didn't really notice any difference with it.

i usually do not add anything.
once per year i will fill the fuel filter on my 88 IDI with ATF start it for 30 seconds and let it sit for 24-48 hours to clean out the injector pump and injectors.
the 02 gets nothing added to it except if it is really cold out in the winter, then i will add a little diesel kleen to keep the fuel from gelling.
12oz of Redline racing two stroke oil per tank. Around 200:1 ratio. I figure if 100:1 can do all the lubrication for a two stroke.. Half that should be doing something good!
Two-strokes run at 40:1 or 50:1. But of course, the fuel is doing ALL the lubricating. 12oz in a tank is actually about 200:1.
To the OP's general question, it's also a matter of your objective. Higher cetane? Anti-gel? Lubricity? Different purposes, different products. I believe it was Jarrett who inspired me to opt for the DK / TCW3 witches' brew.
