The main thing is you'll need a computer and all of the associated wiring to control the E4OD.
Mechanically I think the C6 flex plate will work. The E4OD might be a little longer, so you might need a new crossmember (or move the stock one back a little) and shorten the driveshaft.
Also if the C6 has a fixed yoke and the E4OD has a slip yoke you'll need a new driveshaft.
Then there are the little details like linkages.
Oh, and you don't say if your truck is a 2WD or 4WD. If it's 4WD then you need to convert the 2WD E4OD to a 4WD. The easiest and cheapest way to do that is a two-step process. First convert the 2WD trans to cash by selling it, then convert the cash to a 4WD trans.
