From what i know, no,
however it will screw around with your cruise control and speedometer needle

I know cruise control and speedo won't work but general engine running is what I'm wondering?

I don't see why it would matter. The PCM for an automatic uses the VSS to help determine a shift strategy. You do that part with the manual transmission. I could be wrong, though.
