It would help to know what your truck is. F-150, 250, 350? What engine? What transmission?

Not to hijack but- let's discuss a broad spectrum of ways to improve performance.

1. And most important: Diagnose and repair all mechanical defects and impedaments. If you have bad injectors, melted plugs, shorted wires, clogged air filter, crude viscosity oil, 1/2 a quart of rear diff oil, hanging breaks or warped rotors, bad bearing, Under inflated tires, mismatched gear/tire ratios, extra weight (300lbs of junk and work clothes in your tool box), poor alignment or any other of a hundred regular maintenance projects. A BUNCH of performance comes from the basic ability to PERFORM.

2. Basic upgrades: Highflow exhaust, HF/cold air cleaner, synthetic lube and oils, performance plugs coil and wires, tunes.

3. Hard upgrades: Heads, cam, intake, higher compression, added cubes.

4. Power adders: Turbo, supercharger, N02.

How realistic are 3. and 4. for DD's? I'm considering a SC for my 5.0.

If you want more power out of it, replace that 300 with a 351.

If you aren't willing to do that, maybe look into a gear swap. Find out what gear ratio you have and post it.

What size tires do you have?

Well short of dropping a 351 in there or replacing your 300 with a carb'd 300 horse buildup 300, you're pretty much stuck with a 145 horse Workhorse.

3.55 is a great gear ratio with the 300. 4.10's would give you more oomph around town, but hurt your top end. Just give the ole' girl a tune up.

Nope not really. Basic tune up items. Only real options are carb it or swap motors. An engine swap wouldn't be that hard.

If it were a carb'd 300, you would pull gobs of power out of it.. but not with the EFI 300's.
