You can never have enough tow vehicle, and the V10 gives you the option for moving up in RV. 4500lbs empty could be 6000lbs loaded too. The v10 is the way to go.
I wouldn't hesitate to go with the v10. I have a 2003 E350 with a v10 and use it to tow a 10,000lb travel trailer. I think the 6.8l v10 is the best heavy duty gas motor Ford has ever built.
I get 7-9 MPG pulling a 7,500lb travel trailer. with 350lb bike & (lots of) other random stuff in van.

I have a V8 and it handles a 6500# trailer just fine. As a DD the V8 should get 10-15% better economy but it only translates into 1.5 MPG so unless your annual milage is significant, the V10 won't be a huge extra operating cost.
I have changed the rear end to 4.10. I can normally count on about 8mpg with my 10,000lb trailer. Bucking a wind will lower the mileage. I get about 15 unloaded on the highway. I tow and drive at about 65mph. I also always tow in overdrive unless pulling hard.
I love the idea of a V10 (I have one). The thought of pulling 5 tons (I do it). A dually just to adds to the excitement (yes!) Then I think 40 cents per mile, What was I thinking?!?!?. That baby sits in the driveway now, puts on 30 miles a week going to church on Sundays. My point being for hauling they are great but make sure you have another everyday vehicle for running to the store.
