From my research, I found the frame has the holes for the OEM battery box from the diesels. A quick run to Fleabay netted me two of them for less than $100 w/shipping. I haven't actually installed them yet, but it's a common thing.
Then you'll need some sort of isolator/charger/switchie thingie. I like the Hellroarer Tech version:
Ford factory E-450's and E-550's cut-aways already have twin batteries. Not sure if cut-away E-350's are made anymore. Used for RV's and Ambulances if you didn't already know that.

I thought this was your reason and I too run my van's radio, added interior cargo bay lighting and 12v accessory ports since its an outside work truck. My parts/materials have to be kept warm too during colder months so I did something a bit different---added a RV-type 12 volt converter.
Any device or outlet I wanted to operate that would normally draw power from the battery was wired into the converter in such a way that the chassis battery was taken out of the loop when shore power is applied. There is a 120 VAC heater too but none of it was that difficult to wire together.
All this assumes of course when you're parking and running things you have access to 120 VAC---I do so it was easy to do. You might not have that luxury which makes adding a second battery understandable.
