I just purchased a 2010 F250 xlt. I am a little confused because every local store and tirerack list the truck as having 265/70/R17. However, the truck came with 245/75/R17. This is also what the door sticker recommends. My question is does it matter? Will it affect the speedometer one way or another?
Also the door recommends 80 PSI in the rear and 75 PSI in the fronts. I am about to do a 1000 mile trip with just a duffle bag in the back so what's the best PSI to have in as far as comfort/ gas mileage and be safe without a load?

What will affect the speedo is the overall tire diameter. The speedo is calibrated for the tire size the truck came with. Any change in the overall tire diameter will affect the speedo.
As for PSI to run, what I recommend is the chalk test. Take a piece of sidewalk chalk, and draw a line across the tread of the tire. Drive until the tire makes a few revolutions, then check the wear. If the chalk is worn in the middle of the tread, it's over-inflated. If it wears along the edges, it's under-inflated. If it wears evenly, it's just right.
