Looking for some advise, I have a Inline 6 with a 1 barrel carb. I really don't know that much about engines but i was wondering if i could install a 2 or 4 barrel carb on my inline 6? I know I would have to put on a different intake but would it be worth it? and anything else i would have to do to it?
I just put a Weber 38 Outlaw 2 barrel ( about 420 cfm) on my Chevy 250 six. It made a major improvement in the mid range 2500 3000 rpm and pulls pretty hard, even up hill. It's almost like I have a small v8. I have an Offy intake, so now I'm going to run 3 two barrels and dual exhaust.
It was running lean when I first put it on, so I tuned it with a wideband o2 sensor and had to go up one size on the idle jets. That made a big difference as well.
Good luck on finding a manifold. If you do, you can get a racing holley 2bbl and jet it properly for the flavor of reformulated gas they sell in your area. I find RFG runs way too rich in indiana on historically jetted carburators, fouls plugs and pollutes the oil. I've quit driving my six cyl mustang and have replaced my lawnmower with a diesel tractor because of this. Chain saws and small generators are useless too, the gas here eats up the carb rubber in 1 year. If you find a 2 bbl manifold for a 240 cu in ford 6, please send me a private message with part number and brand, or post it back here.

P.S. I added a set of headers to my old 240 inline 6, man what a differents in ( POWER )!
and thats with a 1 barrel carb, and stock intake! I put the headers on this truck in ( 1986 ) and this guy that I used to work will still talks about how that truck sounds, and how good it ran every time I run in to him, years and years later!
buy the dual plane offy 4-bbl intake headers and toss in a cam. good to go. I would say to try and keep the carb under 500cfm unless your doing head work. I got a 500 carter thats great, but I do need to bring the secondaries down another step.
There are no 2v manifolds available per se. What some do is use an Offy C or Clifford with a 4/2 adapter. The 170/250 small block 6 crowd quite often have the manifold machined to mount a 2v carb as the manifold is part of the head on those engines. The H/W progressive 2v (used on Pintos and Vegas, etc.) is quite popular for the conversion and are readily available.
