rear diff question

I had to put a new ring and pinion in my 86 f250 and after getting everything lined up and set I gave it a test run with the rear end blocked up off the ground. Everything looks and sounded good. I set the gears up with the same shims as my previous setup and that worked out great. The gear teeth wear just like the instruction manual shows. My question is with rear end off the ground should I be able to turn the diff by hand. I am not able to and unit is tight. Truck turns it just fine but I dont want to wreck my diff bearings if there is a problem. Thanks for any help.

Thanks for the reply redneck, yes trans and transfer case were in neutral. Good news though ,I figured it out. I had the pinion nut overtourqued and after fixing that was able to turn by hand. So now we are back together and on the road.
