I installed a pair of fel-pro permatorque gaskets on my engine and I used a thin coating of anti-seize (silver permatex)on the head bolts. I had to remove one of the heads and I noticed that the head gasket soaked up the anti-seize and there were wet spots around the bolts holes. Could this be a sealing problem and/or destroy the head gasket?
if the headbolt in question extends into the cooling passages, a pliable sealant (like loctite PST) must be used or they will leak.
I've heard it reccommended that you use sealant on all the head bolts, so that you get a consistent torque reading among all the bolts. I think you may have hit on the problem quadriver I wouldn't think the antisieze would migrate like that.
It sounds like you're leaking around your coolant passages. Improper bolt tightening sequence, or not "cycling" new fasteners can cause leaks. I use "copper coat" or hylomar spray on anything that has to seal water. The debate surrounding lubes or sealers for bolts or studs has been going on for a long time. For blind holes, that don't penetrate the water jacket, I use only the recommended lube from ARP (which I use exclusively) on threads and under heads and washers. Cycling new fasteners is a MUST. If a bolt or stud requiers a sealer, I use plain old plumber's brush-on pipe dope that I get from the hardware store. Antiseize on head bolts is a no-no, IMO. ARP | Tech - Installation
A warped head, deck, or incorrect finish on the mating surfaces can also cause leaks.
It may sound dumb, but there IS a top and bottom, and a front and rear to the gasket.

Felpro's instructions regarding Permatorque heads gaskets advise against using any coating. I'm glad you mentioned the cycling, I was just at that site yesterday, but didn't go to that part of it. I also was not aware of that procedure and will definately follow that on my next build, even though I haven't had any gasket failures when simply torquing the bolts one time in 4 steps.
