How does the TS compare to the 4R100 and the E40D behind the Power Stroke diesels?
The E40D behind the 460 in my F-250 has done fine for as long as I've had it. I just had the fluid and filter changed in it, and it shifts a heck of a lot better. I got the chance to drive a 6.0/TS this past weekend and it is one smooth operator. The only complaint is that I think it shifts too fast, but I guess its programmed that way for mpg's as well as keeping the 6.0 in its torque band.

The Torqshift does exactly what Ford had it built and designed to do. Work better and handle more than it's predecessors. Shifting fast is actually a good thing. Like you said, it helps get more mpg and more power out of the engine. The more time a vehicle spends trying to shift, the less effecient it is. From what I understand, it's a relatively bulletproof transmission. While I don't own one, I've heard great things about it.
The new torqshift also has the braking feature to help slow you down when going down hills.
It will hold speed on my truck while pulling a 10 klb trailer down a 6% grade and holds it pretty well even on the 8% grades. Nice not to have to ride the brakes down hill.
My '05 F250 with Torgueshift is the first vehicle I've owned that will downshift in cruise control to maintain speed going downhill. The first time it happened it spooked me, but thinking about it later it was just doing it's job. The computer doesn't know if your truck is full or MT. All it knows is maintain that speed. I like it, if you don't want the downshift thrown in, take off the cruise and blast down the hill. They give you a choice.
