My 03 V10 has a vibe upon starting. It is worse when under load or starting to accelerate on a hill or from a 90 degree turn. Was moving its fine just from start to about 10-15 mph. It feels like it may be comming from the back.
What could possible causes be. Any feedback would be appreciated.
I had the exact same issue, most likely its the rear axle limited slip chattering. You could try replacing the fluid and adding friction modifier which didn't work on mine. Because my truck only sees pavement while pulling the camper I replaced the carrier with a open unit and problem solved for me.
First thing I'd check is for any U-joint play by grabbing the driveshaft and seeing if you get any play there. Usually the rear most U-joint at the pinion flange is the one to go first. While your grabbing the driveshaft, check play at the carrier bearing as well.
Jack up the rear axle so the tires can rotate freely and check wheel bearing play/noise as well...

one other question if it is the non slip can I just replace it with a Detroit auto locker? I have them in my jeep and really like em. The ocasional clunking when I put it in drive or reverse doesn't bother me a bit. I would leave the front open dif.

The 2011+ Super Duty's came with an E-locker option. You can buy the carrier with pigtail for about $500. Swap over the ring gear and set the backlash, run a 12V (switched) source and you're done... (Obviously a lot of work but is what I plan on doing in the coming months, hopefully...)
I'm still not sold that it is your differential though (It is a possibility of course). Sounds more like driveshaft related or possibly wheel bearing... but hard to really diagnose without feeling, seeing/hearing it.
From the way he is describing it its identical to the issue I had but its always worthwhile to rule everything else out as I did. I even went so far as to have a brand new drive shaft built for the truck which didn't help anything.
