Difference between front and rear rotor?
With big trucks like these it's never a bad idea to replace all components at once (front and rear rotors, calipers and pads) if you find you need to replace any of the calipers, especially if you find yourself towing equipment or heavy trailers frequently. If you live up north you may also take the time to inspect your brake lines for rust and corrosion. At a bare minimum, be sure to get new rotors and pads if you can't swing getting new calipers on every wheel. Summit has a good caliper kit that includes the brackets so all you do is put new pads in and bolt them on.
There are a ton of how to videos on youtube for replacing calipers and rotors on the 99-04 Super Duty trucks.

You'll want at least two 32oz bottles of DOT 3 or DOT 4 brake fluid on hand. I usually have four of those 32oz bottles just in case.
You'll also want to look at youtube videos on bleeding brakes - you may find it hard to do without a helper to sit in the truck and press the brake pedal - the videos will explain how that works.
