I think without them the tailgate alignment will be off, and the tailgate may rattle.
could you drill through it where the peg is, and then put a plastic wall anchor with a stainless screw and washer through the stop into the wall anchor inserted into the hole. Automotive uses a similar concept for trim pieces using a push pin into an expanding mushroom.
Mine have come out before also. I'll have to look and see if they are still there. If they serve a purpose, they certainly should have a better way to stay put. They do pop out pretty easy.
...I was thinking of using a little good clear silicone to stick them on better?
No, not silicone. That's not a glue. It's a sealer. Get a tube of weatherstrip adhesive. Clean the area with alcohol, follow label instructions. Don't get any on your fingers. Very hard to remove.

Roger that! Need more coffee!!!! Don't know why I said Silicone
My port side tailgate stop/bumper fell off at about 28,000 miles on my 2019 F350. I made an appointment at a dealership, explaining that I wanted it replaced under warranty. I called the parts department first to ensure they had them in stock. They told me the stops/ bumpers are about $11 each. I watched the technician replace the port side one because he did it in the service entrance. He checked the starboard side and said, “I should replace this one also.” Thankfully they had one of those in stock also. I got both replaced at no charge and about an hour of my time because I made the appointment for the first opening of the day. The new ones are stable, and I’m glad to have them. As an added extra, I now know how they are supposed to sit on the truck. I couldn’t figure it out. Turns out they had rotated, and this is why they became loose and eventually fell out.
Too funny... just had my first incident of that thing falling off on mine this weekend. Turns out I only have (had) one on the driver side, no idea when the passenger side vacated the premises. Haven't even looked around yet, but anybody have a part number handy to order them?
