Got my 81 F100 started and running, but I noticed trans fluid running from the front cooler fitting on the transmission. When I was cleaning the fittings up, I accidentally removed what appeared to be an O ring inside the brass fitting. My question is, would the missing O ring be the issue, and how do I correct this? Is that O ring part of the fitting?

Is this near where the fuel line enters the carburetor housing? That's the only place where the fuel is pressurized at the carb. Once the fuel reaches the float chamber, it is no longer under pressure. Same for all the jets and other internal fuel passages. Fuel is drawn through by a vacuum, no pressure there.
I'd carefully dry off the area surrounding the fuel inlet. Look for leaks there. Your engine probably had a fuel filter there. The filter itself may be leaking from a seam and spraying fuel down around the carb base. Or if the filter was removed, the fitting that took its place might be a cobbled affair and doesn't seal properly with the carb's female threads.

I took a video, and I looked back at it, and it is indeed leaking from a seam. Found a replacement trans fitting this morning, have another one on the way to replace the other fitt
You’re going to have to help us out. The mindreading pills available today just don’t work as well as they used to. Most come from China anymore. I miss the good old American-made pills. A guy could take a couple of them and have no problem understanding what you’re seeing right in front of you. Unfortunately, today we are reduced to hoping for an detailed description of the situation.
The preceding was industrial strength satire. I’m just trying to get across that you haven’t given much information. We’ve got to do a lot of guessing to fill in the blanks. Without such details, we can’t really help you. Obviously, you know what you know, but the rest of us are sure at a loss.
You had previously mentioned fuel spraying from a seam. Do you mean from the seam on the fuel filter body? If so, you’ve got a bad filter. Replace it with a different brand and try again.
Or by “seam”, do you mean the junction where the filter threads into the carb body? Or perhaps where the metal line attaches to the inlet side of the filter? Without such details, we are left guessing.
See my pills were telling me he had a 4 cly in that truck of his, could this be true?
So what motor and carb are we looking at?
If it is a V2 2150 or one of the Holley's the accel pump could be leaking and look like it is the body of the carb.

Does your carb have a vent hose on the bowl?
You can just make out the rubber hose on the bowl vent next to the fuel filter.
For my set up it had a solenoid mounted off the carb.
Now being I did not have it I ran the hose to the charcoal canisters and and pull vacuum off it when running.

Can you get a picture of the side where the filter screws in?
I think there is a port opening there with out the pipe nipple to put a hose on but can't see it good.

There is an opening there. I was given another identical carb, that has a mechanical bowl vent setup, I am thinking about having the top, and base from it, put on the carb that I have now, also will give me automatic choke again too.
Sorry I did not see this till now don't know how I missed it.
If you do the top swap just check them both over good to make sure they are the same other than the missing the vent nipple & choke set up.
I can get more pictures of mine but I don't know if it is the right carb for an 81 truck as I have seen the choke hot air tube come in on the other side than mine.

I looked at both tops, they are exactly the same,except the other top has no provision for mounting the choke pulloff, but it has a factory electric choke cap with place for the hot air tube to screw into at the rear like mine has
