There's a seal in the frame to hold it in place yours may be dry rotted and not look like a seal anymore or totally disintegrated. There's a thread here on the replacement of the seal from what others say they are a pain. Here's a link to a new seal.

Guess I might not have explained myself well enough. The vent window glass is actually backing out of its frame. Not the frame that holds the assembly in the Door, but the frame the glass fits into. The bumps locking lever doesn't go through the glass to hold it in place. I've looked through my shop manual but have not seen a reference to what actually hold the glass in place. There has to be something that was used, whether its some type of caulking, tape or whatever. <br />
Anyone know what was used originally to do this?
The glass is held in there with glass tape. You can pick it up at most any auto glass shops around your area. I don't think I've seen any good pics of anyone replacing the tape or frames. If you can post some up as a how to for others.
I am thinking about trying a wire wheel on a dremel to remove what remains in the glass frame. If that doesn't work, will try manual method with a small flat blade screw driver.
I have noticed that my drivers side vent window tries to back out of the frame. Is there some type of moulding or adhesive that holds it in the frame?
The vent glass is retained to the frame with glass tape .. available from any auto glass shop. Glass tape is also used to bond the door glass (21410) to the window channel (21458).
Number dummy, I really do appreciate the exploded view you've provided.
When looking for the window tape I came across this link. While it doesn't show how to remove the old stuff, it does give some interesting tips on the install.

Remove the old glass tape from the frame w/a blade screwdriver. Place the glass tape on the glass, then shove it into the frame.
Once it's set up, remove the excess glass tape with a razor blade, xacto knife or any 'ol knife you happen to have layin' around including an Arkansas Tooth Pick (aka Bowie Knife).
I have more than a couple of Bowie's laying around. That might make for an interesting video...
Texican's would not only know what happened to Jim Bowie, but what occurred across the street at the Menger Hotel bar in 1898. TR met with the Rough Riders candidates.

To be honest, I did not know about the Menger Hotel. Always nice to learn addtional pieces of Texas history. That wasn't something that was taught in the 3 yrs of Texas history we had when I was in school. Thanks for the additional lesson!
Is there a size difference (thickness difference) between the glass tape used for the main door window and the vent window? I have no problem trimming the obviously too wide tape when used in the vent window with a knife, but I want to make sure I am using the correct thickness before I overstuff it all together.
The glass tape I have is what came from LMC. Worked great for the door window, but it "feels" like it might be too thick for the vent window. I've been too afraid to just start stuffing it in there to see just how the fit is - hence why I wanted to ask first.
It would seem so. Today I was able to try and install the vent window glass into the frame and was unable to with the door glass tape for the main door window.
The tape measured at about .056 to .062 (about 1/16 of an inch) and I was able to get it part way, however I could not get it all the way without really forcing it to the point of breaking the glass.
I'm thinking that perhaps 3/64 might be a better fit or perhaps 1/32. I'll have to find a place to get the tape.
Thanks everyone for posting info in this thread. I have the same problem as the OP and while I have the doors apart during paint and new seals all around, I can address this issue as well.
