Searched a bit but didn't find what I was looking for. Have an 06 250 super cab, put a few thousand miles on it and this door thing is driving me nuts. It sounds like it is coming from the internal handle mechanism in either the front door or rear door passenger side, kinda right in the middle.
The doors don't rattle or shake going over bumps, it's a pretty tight seal. The panels don't move either. It's a tick coming from inside that happens over bumps and sometimes it does it for no reason at all. Like a cable wiggling around in there. I pulled the rear panels off and didn't find anything that appeared to be the culprit. Going to do the same on the front door but my gut tells me it's coming from where the door latch/lock mechanism meets eachother when closed.
I thought I read on here somewhere some one posting about it also but could not find it. Thoughts? F-ing driving me nuts. Thanks.
I dont know if it is related at all but someone on here i forget who had a sound coming from thier rear doors as well like you desribed. I think it ended up being a spot weld onthe bottom of the door skin. I'm not sure if it is related but it cant hurt to look i guess.

I've been looking for that noise for a couple of years!
Glad I'm not the only one. My last thought was that plastic latch/lever that sticks out of the rear door, rubbing on the front door. There is a wear mark there - I'm thinking I might tape the latch to see if the noise stops. It seems like it doesn't always happen though...
Dang sucks to hear you have the same thing going on. I suppose it could be worse but it does get me i guess.
I think you are on to something with that lever, I can almost duplicate the sound when I push it in and out, not quite but close. I am going to remove both panels and pull and shake everything I can see tomorrow afternoon. Hopefully I find the culprit I will post my findings.
And yes mine doesn't do it all the time either. It'll do it on a smooth road and then sometimes over decent bumps when you'd think it would tick and it doesn't.
I also have that same tick, I just deal with it although it really bugs me. I could never figure it out the only possible lead I had was the spot weld thing.
Problem not solved but I think I have found the culprit. Today I started with removing the front door panel because I already did the rear door and nothing changed.
I had added some weather stripping I had laying around to some of the latch and lock mechanisms inside that seemed to be a little loose. After doing this I drove around a little to see if it helped which it seemed to(may be wishful thinking) but the noise is absolutely coming from inside the front door. It was much more audible with the door panel off and the plastic insulation pulled back. I am almost certain that the latch mechanism is the culprit, something inside because pretty much everything outside doesn't have a significant amount of movement.
I started to take it apart via the three torx screws around the latch itself(not in pic) and couldn't really see a good way to get it apart safely. I will try to locate the part and retry in case I break something.
So, what would be the best way to locate this part? I suppose tousley(sp?) would be a good start i will do that and see what I can find.
Hmm I braced around it with the weather stripping and it still made the noise. I will have to look into it though because that thing does move around more than anything else in there. I hope it's that easy. Hence the "almost certain I found it" lol.
My '04 does it if I have both driver's side windows down at freeway speed, like on a hot summer day. It's irritating as hell, but I just pull rank and make my son roll up the rear window.
Figured I would update, I just pulled the panel back off and removed the door lock actuator rod completely and it still makes the noise. So it was not a fix for me but thank you Super Duty Scaler for the tip that is why I posted.
