I have the E4OD in my 2000 EB Expy. I do haul a 16' car hauler with a five thousand pound Genie scissor lift with it.
I am curious to know if one transmission is better than the other for future reference when this E4OD tranny goes.

4r100 & the e4od are basically the same trany. the 4r100 is an updated e4od and made to bolt up to the modular motors. the 4r70w/4r75w are smaller tranys and are an updated aod/aode and made to bolt up to the modular engine family. the 4r100 is the same trans put behind a diesel and very relible when taken care of.

Your 2000 Expy already has the 4R100 if it is a 5.4L. They stopped making the the E4OD after 1998.
I wouldn't expect the trans to give for a long time unless you are not caring for it. I got a solid 200k out of a 4R100 behind a 7.3 deisel in a '99 F350. That truck was either pulling 15k lbs or plowing a 10 acre yard in northern Minnesota all its life. It hadn't even failed at 200k but was rebuilt as preventative maintenance. It did get 30k trans fluid flushes and filters though.
